Can AI screen which chemicals biodegrade readily? External data sharpen the claim

A reproducible model reached external ROC-AUC 0.911 across 670 compounds, supporting prioritisation for testing—not replacement of OECD biodegradability experiments.

The short version

What happened
The benchmark separated 837 compounds for training, 218 for internal testing and 670 for external validation, rather than relying only on cross-validation.
Who it affects
Faster prioritisation could direct laboratory capacity towards uncertain chemicals, but false reassurance would carry environmental and public-health costs.
What remains unproven
Labels inherit benchmark quality and coverage

Keep in mind

External validation was retrospective

Readily biodegradable recall was 0.754

A held-out external set tested more than model fit

Researchers in Saudi Arabia and Egypt used molecular descriptors and fingerprints to classify chemicals as readily or not readily biodegradable. Data were divided into 837 training compounds, 218 internal-test compounds and 670 external-validation compounds. Feature screening reduced 119 RDKit descriptors and 1,024 Morgan fingerprint bits to 311 predictors before comparing four model families.

Tree ensembles performed best. Recalculation from deposited gradient-boosting predictions gave external ROC-AUC 0.911, accuracy 0.866, readily-biodegradable recall 0.754 and Matthews correlation 0.668. A paired bootstrap put the external ROC-AUC 95% interval at 0.886–0.935.[1]

Useful triage, not a substitute for environmental testing

The workflow uses open tools and includes applicability-domain checks plus global and local explanations. Surface-property and topology descriptors contributed strongly, while individual hashed fingerprint bits are difficult to interpret chemically. The practical use is ranking compounds for testing or auditing a model-assisted decision.

A prediction does not measure degradation in water, soil or treatment systems and cannot establish a causal mechanism. Roughly one quarter of readily biodegradable compounds were missed at the reported operating point. Prospective validation on newly tested industrial chemicals is the next test.[1]

Can AI flag chemicals that bind a hormone-carrying protein? An FDA-led model found useful signal—but only nine external non-binders

A consensus of four molecular models reached 0.430 Matthews correlation on an external set of 164 chemicals. Its 88.4% accuracy is easy to overread: 155 external records were binders, seven came from non-human assays and the model has not replaced laboratory testing or predicted harm in people.
